Dell PowerEdge R510 with two Intel Xeon E5645 (6-core) CPU's.

Windows Server 2008 R2 with Hyper-V role installed.

5 Virtual machines, four of which will need backup and restore ability at the file level (one vm is running Lotus Domino server).

IMO.. This is what you will need..

  1. Backup Exec core license = One
  2. Agent for Hyper-V = One
The Symantec Backup Exec Agent for Microsoft Hyper-V lets you back up and restore all virtual machines that reside on the Hyper-V hosts.
Backup Exec's Granular Recovery Technology (GRT) is enabled by default for backup jobs. You can use a GRT-enabled backup to restore individual files and folders from a Windows virtual machine without restoring the entire virtual machine.
In case you need more detailed information, you can get in tough with the licensing team:

Thanks for the quick response!

Dell recommend that I purchase..

1 x Backup Exec 2012 Virtual Server Suite

2 x Backup Exec 2012 V-Ray Virtualisation Edition 2-6 core per Socket

4 x Backup Exec 2012 Agent for Windows.

It's the V-Ray (x2) license that I'm not sure about and when I ask Dell, they're not sure either but recommend that it's required.

I've just emailed the licensing team with the same query.
