Open an Agent, click Settings then Update Configuration.
if the Last Configuration Date changes then Communication is working.

Also, if you've observed that Agent doesn't communicate with NS Server, then first you can ensure that client PC is online and accessible from NS Server, using mouse right click menu to ping it from Console
After that you can open Resource Manager of client PC to determine whether there were crashes of Agent, etc.
Also you can check client SMA state, using Real Time System Management solution
