I am currently running Symantec Endpoint Protection on several servers all of which are running Windows Server 2008R2.
    SEP version number is 12.1.4112.4156.

Data files are currently being stored in two locations:
    C:\ProgramData\Symantec\Symantec Endpoint Protection\12.1.4112.4156.105\Data\Definitions\VirusDefs

    and

    C:\Users\All Users\Symantec\Symantec Endpoint Protection\12.1.4112.4156.105\Data\Definitions\VirusDefs

Is it necessary to have the same data in both locations.  Is it possible to remove the files from one of these locations without affecting the Symantec Endpoint Protection client?  If so which location can I remove?  Current size of each VirusDef's folder is 1.6 gb.

It's a junction point, see SMLatCST's post here:

    Really only takes up 1.6GB
