I have a problem with the GUP's configuration on SEP 12.1. In particular seems that GUP works only it I define a single client that works like GUP:

but if I define a multiple GUP list that contains also the IP that I have indicated in the previous configuration like single GUP IP, GUP doesn't work and in the client's appears this message: "Failed to connect to all GUPs, now trying to connect SEPM"

I have already checked GUP configuration on SEPM, and SEPM recognizes correctly the machines that I have indicated like GUP (Group Update Provider=True).

on SEPM in installed the SEP 12.1.4104.4130, on GUP the SEP 12.1.4013.4013, while the SEP version install on clients is the 12.1.xxxx.xxxx.

Your SEPM Server and all GUP clients are similar version?
