This issue has been solved. See solution.

In my organistaion there are few comuters( unmanaged client )  which are not in any kind of network & its used for some testing purpose, though I installed intelligent updater version of updates, its not updating proactive threat protection & network trhreat protection, I tried uninstalling SEP but still the same issue

Hi,
Intelligent updater will only update the definitions for the AV/AS component. PTP and NTP definitions can be updated onnly from the liveupdate server of internet, or Liveupdate Admistrator.

So, I suggest you install an LUA in that network and attach the clients to LUA  for getting the definitions.

But at the LUA will need to have internet connection to update the virus definitions.

Please following documents for Installing and Configuring LUA.

Also, plese take a look at the following Discussion Forums and its solution for connecting the unmanaged clients to the LUA.
