you can use the jdb file to SEPM manually. Note that it only updates AV definition.
You can also configure the SEPM to get the updates from LUA ( which will be downloading definition from internet).

Live Update Administrator
Live Update Administrator (LUA) is what you need in your case. This software is able to download the definitions for the Symantec products you need. Then, Symantec Endpoint Protection Manager can be configured to download the definitions from LUA.
Check this:

What are the Symantec Endpoint Protection (SEP) versions released officially?

Hi, You have two options
Hi,
You have two options i.e LUA or Manual update.
With manual update you have limitation becuase it will update only AV definitions.
I would suggest go with Live update administrator.

JDB or LUA for different purposes
I agree with the above posters that you should use either JDB files or LUA.
If it is sufficient to update your SEPM only with Antivirus/Antispyware content (i.e., no content for IPS, TruScan etc), take JDB files.
If you want the complete content and your SEPM has the permission to access a different network, install a LUA in the other network (which has access to internet) and update the SEPM through the LUA.
If you want the complete content and your SEPM is in a high security network (no access to other networks), install a LUA in a different network, create a distribution point (webserver or share) in the SEPM network and copy the content from the LUA to the distribution point in the SEPM network. Then update the SEPM from this distribution point. See here: http://www.symantec.com/docs/HOWTO44060

You can support an idea to integrate all kinds of content in JDB files here:

Hi eliteeagles,
If you don't want to update manually then go with Enterprise Edition.
You can upgrade from SBE 12.1 to Enterprise Edition 12.1
