This issue has been solved. See solution.

We have a company where there are at least 25 sites, and not ALL newly imaged machines are going to be in the same place. I will list 4 sites.

New York

Los Angeles

Houston

Chicago.

I don't want them ALL on the same site, what I want is for them to report to their OWN GUP server.

What do I need to do to get Chicago clients to point to Chicago GUP's
New York to New York and so forth...

The machines will come here and Iwant to have these machines with SEP already installed. I have a package, but it will point them to a specific group. ALL of them wiill default to the New York group based on the package, what I want to do is to divide them accordingly, and report to their servers when they get to their respective sites.

The GUP's are not in place yet, but when they are, I want the clients to point to them. What is the best approach to  take at this time?

With RU5 multiple GUPS can be

With RU5 multiple GUPS can be configured for any group and the RU5 clients will parse the group list and only use GUPs in their own subnet, by IP address and subnet mask; So in that respect your job is relatively easy. Just make sure each city has one (or more) GUPs for each subnet.

Adding to what Jeremy has

Adding to what Jeremy has said with RU5 it is no more  a GUP for  a group , it is GUP's as per the LU policy.
The  GUP will update all the clients in its Subnet.

If you NY clients are in the same subnet and your Chicago clients  are diffrent subnet they will go to their respective GUPs.
