In Sep 12.x Tamper protection are enable by default you can  disabled Tamper Protection.

How to Disable "Tamper Protection" on a SEP 12.1 Client

In addition to files being protected, SEP 12.1 tamper protection now protects the registry as well. You will need to disable tamper protection in order to make changes.
