yes. First, the client will contact SEPM. SEPM will build the definitions and make it accessible in IIS mgr, it will send client, the URL for the published content.

Then the client will contact GUP, asking for that URL, GUP , then will download the content from SEPM and save it in the sharedupdates folder. GUP will provide the client with the definitions it has received from SEPM.

So, YES, GUP can update clients that do have PTP & NTP installed despite of its own installed components.
