I am re-structuring how our clients receive AV defs from GUPs. Where do I specify that Mac OS clients receive definition from GUPs. Some of our branches have 90% Mac OS computers.

Mac clients cannot receive content updates from GUPs. this is only for Windows clients. See the Mac FAQ here:

A. No, for the same reasons outlined above.

As of SEP version 12.1 RU4 the SEPM can be configured as a reverse proxy for downloading and caching the latest Macintosh LiveUpdate content. All Macintosh updates otherwise must otherwise occur through LiveUpdate, either from Symantec's servers or from an internal LiveUpdate server using LiveUpdate Administrator (LUA).

Brian is correct.  With SEP 12.1.4, Mac clients can receive their definitions from the SEPM: here are a few articles with the details....

Be aware that this process allows the SEPM computer to cache Mac content.  It does not process those Mac definitions into .dax files the same way that it can for Windows clients.

Yes only Widows clients will fetch the updates from GUP. Mac client will fetch from SEPM or Symantec LiveUpdate.
