No..Its not
No..Its not mandatory..however its a best practise for the clients..so you can reboot anytime after the install may at the weekend or so..however its not required.

For SEPM no reboot required atall.

@ zenworksb : No, reboot does not need when upgrade to mr5. Just select group of user you want to upgrade, and woalla...your client version changes from mr4 to mr5..
