Have you tried to reas through the NBU for VMWare guide yet? http://www.symantec.com/docs/DOC5162

You will find requirements and detailed howto steps.

The NBU for VMware Admin Guide that Marianne mentioned is a must read.

I also highly recommend to read Abdul Rasheed's "Nuts and Bolts in NetBackup for VMware" on the NetBackup blog:

Have you seen the following in the Introduction chapter?

NetBackup for VMware does the following:
■ ....
■ ....
■ Backs up the full virtual machine.
■ Backs up the virtual machines even when they are turned off.
■ Can restore selected files from the backup.

"full virtual machine" is the vmdk.....

You can not back up VMware files in vmfs directly.
Only way we have allowed is to backup VMs via VADP using NetBackup for VMware.

As stated the direct backup of vmdk files is not supported

All backups should be done using the vSphere API for which you will need a suitable VMWare backup host

If your backup go directly to tape then i would suggest adding a Windows Media Server that shares your tape drives and also has fibre mapping to your ESX Datastores

Once this is done you will be able to configure the credentials and a VMWare backup policy

The Admin Guide covers this pretty well

Hope this helps
