The previously existing option for backup gups is no longer existing in SEPM 12.1 RU2 and has been replaced by Explicit GUP functionality.

Backup GUPS as per description:

"Backup" Group Update Provider on a different subnet (if Group Update Providers on the local subnet are unavailable)"

...were allowing for specification of a GUP machine out of the local subnet, that would be asked for updates in case other gups are no longer available.

In RU2 this option is no more. Explicit GUP settings allows only for a strict assignement of subnet clients group to GUP or GUP subnets, and does not allow any backup options like before. In environment with sub/supernetting backup option was really important.

Would be welcome to see the backup option back alongside the explicit GUP functionality.

i guess you can still achieve it, however it is good to have. you can use the Multiple GUP along with the single GUP which make its the same. Since the sequence it is processed is multiple GUP and then single GUP. backup GUP can be put as single GUP.

...as far as my understanding goes both single gup and multiple gups from the policy will be put in the same globallist.xml - from the client perspective it won't be possible to see which one was set as multiple which one as single.

What I mean with that - are you sure if in that configuration any of these gups will be accepted if it is outside the local subnet?

though haven't tested ...with the logic of processing order, it should check outside GUP i.e. single GUP if the GUP in multiple subnet is not available.
