Looks like Symantec is on the roll - they have just recently released a new update - SEPM 12.1 RU6 MP6 (v12.1.7061.6600) - hurrah!
The following details will get you started with upgrading to the latest version and some other details, but first; a quick summary of what's new:

I would like to upgrade from 12.1.4104.4130 to 12.1.6MP6.
My question is: After downloading 12.1.6 MP6, can i directly execute setup file in existing running environment after stopping manager service.
