Suppose if i have 1 application and i have define proxy in it.

Suppose that traffic pass traffic on random port like 9443 apart from HTTP or HTTPS so in what mechanism proxy understand it in Explicit deployment.

               In an explicit proxy deployment, the client browser will hit the proxy on a defined port and pass the url it is trying to connect to. If it is a url with non-standard port like 9443, proxy will be attempting a detect protcol on the same to identify the underlying protocol. If it is not able to or DP is disabled, it will tunnel the connection as usual.
