What version of SEPM are they running and are these SQL DBs? Have any changes been made recently that would cause communication to stop? Can they both successfully reach each other via ICMP? You may want to start with the replication troubleshooting flowchart:

Check the system times and make sure they match. I have seen communication between 2 SEPMs fail due to the computer time drifting on one of them.

see if you have firewall in place

Firewall session timeout causes Symantec Endpoint Protection Manager replication failure

try rebuilding the indexes, its under admin-servers -site - "Rebuild the index now"

    Rebuild Indexes

To improve the connection time between the database and Symantec Endpoint Protection Manager, the management server rebuilds the database index at a scheduled time. You can perform this task manually at any time by clicking Run.

You can also change the time for when the management server is scheduled to perform this task.

Symantec Endpoint Protection Manager replications time out after several weeks
