When antivirus is running on the servers, do you need to exclude any directories or processes from on-access and on-demand AV scanning on Enforce/OracleDB servers and detection servers?

if you are using DLP agent and SEP agent then you can check this article to exclude http://www.symantec.com/connect/articles/exclude-sep-agent-dlp-endpoint-agent-configuration If SEP and oracle server use this link to exclude the oracle folders and files http://www.symantec.com/docs/TECH134383

We are not using SEP and that info was backwards to what I was asking.

I wanted to know if there needs to be exclusions added to third party antivirus software to not scan certain DLP files.

Maybe I need both exclusions in the DLP agent to not scan antivirus files and exclusions in the antivirus to not scan frequently used DLP executables or folders.

Are there any DLP files that should not be scanned by AV, or does it handle AV scanning with no issues?

for any AV you can exclude the oracle folders as in the link. if you are using end point agents, follow the article however instead of SEP exclude the AV that is being used at your place.
